Microsoft to Retire Some Zune Pass Features

Microsoft is making some adjustments to Zune before the Xbox Music debut on Friday. Gone will be the Mixview playback and channel playlists, direct purchase of stream music lists and a host of other features that will effectively signal the end of Zune.

Microsoft has all but killed off Zune, with a future desktop replacement set for Windows Phone 8 and a new Xbox Music service launching shortly.

a few of my favorite things: preparing

Well, we've got just over 5 weeks before D-day (due date, that is:). Physically, I'm more than ready for this little guy's arrival. Practically speaking, not so much. A lot has been happening around here to try to get our house and family prepared. Here are just a few of the goings on:

1. "practicing"
Anna really isn't much of a baby doll person, but we pulled out the one she has so we could start "practicing". We made a big deal of getting him dressed in some newborn clothes and she's been working on swaddling him. She's pretty excited about baby Knox and I think she's going to be super helpful when he comes. (That's her shy face below) Owen is the one I'm more concerned about. He is very much a mama's boy and doesn't do well when he's not getting my attention. I think he's in for a rude awakening. Hence, the attempts to prepare him by talking about the baby doll.

2. Baby gear and house prep

I was in Middletown a few weekends ago for a baby shower and brought home most of our baby "gear" that was being stored in my Mother-in-law's basement. I immediately felt so much more prepared once I had a bag of newborn clothes and a bassinet. Not to mention bouncers, a swing, a bathtub, etc.?

I've never had a newborn in a two story house before, so that has posed some interesting logistical questions, particularly since our room is on the first floor. If your house is set up this way, I'd love to hear how you best set up house when bringing home baby. We'll have the bassinet in our room for a while, so that will eliminate running upstairs to the nursery multiple times a night, at least for a while.?

I'd been hunting for a reasonably priced dresser on Craigs List for several weeks that I could use on the first floor as a changing station and "keeper of baby stuff." I was frustrated after missing the sale on several pieces and decided to try Trader's World (flea market) with my mom while we were in town a few weeks ago. We came across this piece and I immediately loved it. Great shape and didn't need repainted or refinished. Woohoo!

3. Doctor's appointments
Since moving 10+ months ago, we haven't really switched over all of our doctors. Man, it's hard work finding all new doctors, checking on insurance, and filling out paperwork! I decided that it would probably be best to get in as many appointments in a) before Knox's arrival (when leaving 2 kids with a sitter is easier than 3) and b) before the end of the year. This week I went to the eye doctor, the dentist, and my regular OB visit. Phew. And next week Anna goes to the doctor for her (very late) 3 year check up so she can get cleared for preschool. Which leads to the next point of baby prep/transition.

4. Preschool and potty training
We decided to enroll Anna into preschool a few mornings a week. I think that it'll be really good for her to have something regular to go to outside the house and without me. It will also give me a little bit more time with just Owen as he adjusts to sharing me with Knox. That's our hope anyway! Anna is really excited about school and it was another good incentive for her to really master potty training. She's been doing great with #1, but has refused to go #2 on the potty. She would just wait until we put her in a diaper before bed or she'd ask for one at naps. Today she went for the 2nd time (without prompting!!) and she was SO proud of herself. Almost as proud as me:) Here she is enjoying the reward we've been promising her for weeks. I love the look of satisfaction.

5.?D-Day plan
The last thing that had been kind of stressing me out was putting a delivery day plan into place. Moving away from all of our family made this more difficult than I thought. When I went into labor with Owen we just dropped Anna with our sister-in-law who stayed at home with our nephews and lived 6 houses away. We finally have a "day plan" and an "evening plan" in place thanks to two wonderful friends and I feel SO much better knowing that we have these people on-call to help us out with Owen and Anna until my mom arrives for relief. Seriously, thank you!!

My emotions teeter back and forth between hopeful anticipation and sheer terror/anxiety as we await the arrival of this sweet boy. I'm really so excited to finally meet him and potentially complete our family. On the other hand, I don't trust my own emotions and how easily overwhelmed I can get and I worry about how it's all going to play out after we bring him home. Pray with me that the Lord will prepare my heart, our home, and our family and that I will have peace and trust that he will provide us with everything we truly need. Thanks for your support, friends!

Ex-Stock Broker: I Realized That Most Of What I ... - Yahoo! Finance

Follow The Daily Ticker on Facebook!

Many of those who work on Wall Street go through a process in which they gradually learn that what is perceived as "smart investing" is often unbelievably dumb.

Specifically, they learn that many of the recommendations that Wall Street makes--and the transactions that Wall Street gets paid to facilitate--are not in their clients' best interests.

And once they learn that, they face a choice:

Continue to make the same bad recommendations and trades

Or change the way they do business, often in exchange for a lower initial salary.

One of those who changed is Josh Brown, a former stock broker who now writes a well-read financial blog called The Reformed Broker.

Once he realized that he was hurting clients by selling them stocks he didn't think they should buy, Brown abandoned being a "broker" and became a financial adviser at Fusion Analytics. In this new job, Brown says, his interests and his client interests are more closely aligned.

Most people don't recognize that there's even a difference between a "stock broker" and a "financial advisor," but there is. And for clients, it's a critically important difference.

Stock brokers. who generally work for brokerage firms, do not have to recommend trades that they think are in your best interests. Rather, they just have to recommend securities that are "suitable" for you (a much lower standard). Financial advisors, meanwhile, have a fiduciary duty to act in the best interests of their clients. Although even this standard is often violated (by financial advisors who don't understand the importance of low fees to investment returns, for example), it's a much higher standard. And, all else being equal, no one looking for sound financial advice should ever work with a stock broker over a financial advisor.

Four Steps You Need Before Self Publishing Your Book | Alice Osborn

Pen On BooksSelf publishing your book can be overwhelming, but if you think of your process in Four Easy Steps, you?ll be able to get through and even enjoy the journey!

Marketing/Publicity

?When I was working on self publishing?Right Lane Ends (Catawba, 2006) six years ago, I booked my book launch (Fri. Oct 13th) before I even signed the contract for publishing the book! I had the end in mind and worked backwards to get my book in my readers? hands. That?s what I want you to do when you?re planning on self publishing?what kind of marketing strategy do YOU have so readers can get a hold of your book. Book club tours? Readings? Wine tastings? Charity events? You need to have a marketing game plan BEFORE the book is unleashed unto the world!

First of all, I highly recommend you all buy this book:

The Essential Guide to Getting Your Book Published (Workman Publishing, 2010) $15.95

By Arielle Eckstut and David Henry Sterry

It is comprehensive and is for both traditionally and self-published authors.

?

Website

Your home page should feature your book along with a clear way for visitors to buy your book. Tell them why they should buy your book! What does it give them that is not currently out there on the market?

?

Your website should contain

  • Samples of your writing, your bio, photos from the present and from when you were a kid to create interest, a calendar of your events, open mics, networking, shows, workshops, speaking engagements, etc.
  • A resource section of your favorite books and your favorite links
  • Videos and your book trailer
  • A future projects section
  • Contact info

?

You?ll also need to think about what colors you want to website to contain and how it should look. I recommend you check out and research other authors? sites and print out their pages/take notes to see what you like. I did exactly this when I first designed my website 6 years ago.

?

You can choose whether your book title should be your URL or it should be your name. Experts are divided either way. I use aliceosborn.com since I want to showcase my entire brand and platform

?

Many authors are using WordPress (http://wordpress.com) either as a free blogging platform or they?re using a WordPress template that is running on a domain name they pay for each month.

?

I recommend you use a WordPress expert or a website designer and then have them show you how to implement updates on your own. You never want to call a webmaster (that?s not you) to add events on your event page.

?

Publicity

?

Set a publication date and build excitement around it! You should use this pub date in all of your marketing materials and in your press releases. Talking up your book via pub date in social media is a lot easier than just talking about your book.

?

On Facebook either on your Profile Page (the non business page) or your Business Page, I?d set up an event to talk up your book launch and publication date and then invite your friends to it build excitement.

?

The Social Media you need to use includes

?

Facebook ?you can have a virtual tour on Facebook

Twitter
Pinterest

LinkedIn

Google +

?

You Tube with instruction videos/writing motivational video and video trailers

Blogging

Blogging tours?approach bloggers who review books. You go write on other sites during your tour

Once you?ve become known as a person who is generous, comments on others?

posts and gives out great info on her blog
(For ex: you could blog about why it?s important to exercise as a writer or what books you?d recommend for new writers) your friends/followers will be more likely to spread your message about your new book?s launch on your publication date!

?

Book Signings/Readings

?

  • Use a press release (more press release and press kit information is in the other handout)
  • Ask local booksellers to host you? (you?ll have to put your book on consignment, usually 60/40 retail split)
Also think about having your book launch at a coffee shop, a deli or an art gallery!
  • Get a poster made of you holding your book
  • Approach book clubs via Meetup.com
  • Get your book into the library!

?

Cover Design

?

Your cover will need:

  • ISBN #
  • Figure out your book?s category: fiction, memoir, cookbook, poetry
  • Grab blurbs
  • Cover Art?spend money on a good photo/art/cover designer?this is your book?s primary marketing tool!
  • Bio (max, 150 words)
  • Author Photo?spend money on a good photo!

?

Pay attention to your cover! It?s your most important marketing piece and is what your potential readers will see first!

?

Hire a cover designer to work on your cover or your book designer. You can also find a good stock photo on sites such as http://www.photoxpress.com or http://istockphoto.com

?

Use bright colors in your cover to catch reader attention. Bright colors include yellow, red, purple and blue. Don?t use green?it blends in too much. Try to use white type since you can see white type better in dim light than black type.

?

Four color covers will be more expensive to print so consider a two color cover (black and red/green and white)

?

Make your spine stand out! Always have the identifiers of your name and title on the spine and if your book is wide enough you can also include a photo on the spine. If you can?t have a photo on the spine consider color blocking and perhaps scroll work.

?

Test market your cover choices with your friends/followers/Facebook friends!

?

?

Layout

Hire a book designer to help you with layout and the template. Word is not a good book design software tool!

?

Ask what should my chapter headers look like? Are you going for an ornate or simple look?

?

Standard book sizes are 5.5 x 8.5 for a trade paperback and 6 x 9 for a hardback. Anything outside the standard box sizes will cost you more money. These books are perfect bound, meaning that they have spines like a regular book. Saddle stitched means staples down the center and no type can be placed on the spine. With your book designer, you can talk about the ?trim size? of the book?the finished cover and page size?along with paper color and weight, and number of colors to be used on the cover and the inside pages.

?

The more pages you have, the more it will cost you in both printing costs and in weight.

?

What kind of paper? Uncoated book stock is the best choice, but if you have colored illustrations consider coated papers.

?

Make sure your page color is off-white. Stark white is a dead giveaway that your book is self-published.

?

Many POD (Print on Demand Companies) offer basic templates and I suggest that you study both traditional and self published books in your genre (poetry, fiction, how-to nonfiction, memoir) to see how their pages and chapters are set up.

?

When you?re setting up your interior be aware of what font you want (most books are 10-12 font). Also be aware of the amount of white space in between paragraphs and the amount of spacing between lines (the leading).

?

Editing

?

If you?re serious about publication, you?ll need to hire an editor before you send your manuscript off to agents. If you?re self publishing you also need an editor so you?re work is the best it can possibly be. A good editor will help you proof your work so those contractions and punctuation mistakes are no more. She?ll help you with clarity and consistency.

?

She?ll make sure you keep your book reader-centered, will remind you about timelines, will keep your plot in focus and will suggest new ideas and story directions to make your book better so you will be thought of as an expert. By doing so, your book will be able to help others because it will be read! Sometimes the greatest compliment a self-published author hears is, ?Wow your book doesn?t look self-published!?

?

Even after you?re done editing, you should take your book to a proofreader (who?s not the editor) to catch any additional mistakes.

That?s all, folks! Good luck!

A leaked report in Mexico affirms that the US officials attacked by Mexican Federal Police last week were CIA agents, something that has since been confirmed to InSight Crime.

Writing for El Universal, Mexican journalist Carlos Loret de Mola claimed on August 28 that he had been privy to a confidential report confirming that the two US officials injured after Mexican Federal Police opened fire on their armored Toyota SUV in Morelos state on August 24 were in fact Central Intelligence Agency (CIA) officials.

A high level intelligence source has since confirmed to InSight Crime that the men were CIA personnel.

The document seen by Loret de Mola is reportedly based on testimony by the injured agents, who were supervising a training course for Mexican Navy officers at a base in Xalatlaco, Mexico State, near where the attack took place. It confirms that five vehicles were involved in the attack instead of four, contrary to initial reports.

InSight Crime Analysis

Notably, while US agencies the Federal Bureau of Investigations (FBI), the Drug Enforcement Administration (DEA), the Pentagon, and Immigration and Customs Enforcement (ICE) all denied that their personnel was among those injured, a CIA spokesowman did not issue an outright denial, and instead referred inquiries to the State Department, reported the Washington Post. These latest revelations also contradict a Proceso report based on an anonymous source that claimed the injured agents were DEA.

CIA presence in Mexico increased last year after the US deployed more agents south of the border to work alongside Mexican military officials in the fight against drug trafficking organizations.

The fact that the injured agents were CIA could foster more conspiracy theories about why and how the attack took place the way it did. The incident is already likely to be highly scrutinized since it is clearly an embarrassing episode for the Mexican government, and has (unsurprisingly) prompted President Felipe Calderon to issue a public apology. The US ambassador to Mexico, Anthony Wayne, has announced that the US will work with Mexico to investigate the 12 federal police currently being held for their part in the attack.

The Confucius Institute at the Ateneo de Manila University, in cooperation with the International Society for the Studies of Chinese Overseas (ISSCO), held an international conference entitled Chinese Language Teaching and Education in a Globalizing Southeast Asia last August 17-18, 2012 at the Crown Plaza Galleria, Metro Manila.

?

Topics discussed were on Chinese Language Teaching and Education in Southeast Asia history and politics; culture and society; strategy, promotion, and marketing; methodology and policies; and challenges, among others. Presenters came from Malaysia, Singapore, Indonesia, the United States, as well as the Philippines. 40 teachers from both the Confucius Institute and Chinese schools in Manila were invited.

?

The conference featured Southeast Asia regional characteristics, statistics analysis, and topic oriented sessions. For the regional characteristics, 14 presenters from different countries introduced different situations like whether the Chinese education in Singapore belongs to ?Hua Wen Jiao Yu? or ?Hua Wen Jiao Xue?; why in Malaysia the Chinese education is considered as a mother language rather than as a second language. Secondly is the statistical analysis which made the speeches more persuasive. Such as on the first session of the conference, Professor Leo Suryadinata, Director of the Chinese Heritage Centre; Professor of Rajaratnam School of International Studies, NTU, gave a speech entitled Recent Chinese Language Education in Southeast Asia: Some Reflections in which he presented a brief analysis of Chinese language education among Southeast Asian countries. In the topic oriented sessions, 14 presenters were divided into 5 major topics such as history and politics and culture and society. This layout made the sessions clear and concise in their objectives.

?

Throughout the conference, the presenters elicited comparative lessons, presented new insights and inputs, and tackled current research on how Southeast Asian countries have addressed the challenges of supporting, maintaining, and promoting Chinese education and teaching. What was presented to the audience was a high-specification, high-level international Chinese Education Conference.

?

After the meeting, the organizers will publish the proceedings of the meeting in both Chinese and English languages to let more people know about Southeast Asia Chinese teaching and Chinese education and essentially let the results of the seminar be widely publicized.

You've Decided to Rent an Office ? What's Next? | Work At Home ...

You have a business and have decided that you want to rent an office. Seems like this would be a pretty simple procedure but there is a lot more to consider than just strolling through the ads in the paper and taking a look at some building spaces. If you want to find the right kind of office space, you have to put a little more time and effort into the process to ensure you end up renting the right space for your needs.
Many owners of businesses jump into the first office space they find because of the fancy layout or the luxury amenities. Before you go looking for a space to rent, write down everything you need and take it with you. If you have 5 employees, don?t rent a space for 50 employees. If you are planning on expanding at some point, a little extra space can be used, but its best to rent for current office needs.

Anyone can find office space for rent on their own, but it?s such a big decision to make and will be worth the commission you have to pay a real estate broker who has a specialty in renting commercial property who can guide you through the decision making process that will be best for your office circumstances. A real estate broker can help negotiate the terms of a rental agreement and will have much more availability to rental properties that may not be listed anywhere you can find them.

Have a budget in mind before you go into a rental agreement. Don?t get yourself over-extended because you can make upgrades later on when you?re ready. Office space rental can really be something that makes or breaks a budget. Find a good space that is enough for you to work but also within your budget range. Always take the time to stop and read the small print on the contract. You need to be aware of the extra costs such as utilities or any hidden fees that may be a part of the fine print. This is another area where an agent can come in handle and make sure your signing something that is in your best interest.

Another area an agent will come in handy is with negotiating your monthly payment. Don?t just accept what someone offers you the first time. Use some of the market knowledge you have or just let your experienced broker handle making a counteroffer. Often times, an agent can negotiate because they know more about what others in the area are renting for. Utilities and such can be negotiated as well with the landlord. Some landlords will lower your payments for a longer term lease, and some will trade out part of your rent for your services. This is where a good broker can come in handy to help you negotiate.

You want to be sure your office rental agreement is taking your future needs into consideration. Make sure the agreement states if you can remodel the space or expand if you need to. What happens if you need to end the contract for some reason? You want to be sure of the fees involved with all of this before you sign the agreement.

Slain dictator's daughter turns to mother's legacy in bid to lead South Korea

OKCHEON, South Korea (Reuters) - Park Geun-hye was 22 years old when she washed the blood from her assassinated mother's dress. Five years later, she recalls in her autobiography, she held her father's blood-soaked shirt after the South Korean strongman was shot dead.

Now, both hampered and helped by the contrasting legacies of her murdered parents, the 60-year-old Park appears on the cusp of becoming South Korea's first female president.

Park was chosen as presidential candidate for the ruling conservatives last week and polls show she is the front-runner for a December election.

For some South Koreans, it is the memory of Park's father, Park Chung-hee, that comes to mind when they think of her. He was a military dictator whose 18-year rule dragged the country out of poverty but at the cost of human rights.

He is still a controversial figure in the now-prosperous Asian industrial power. Park's opponents will aim to tar her with his brush in the election race.

But it is recollections of her mother, Yuk Young-soo, once known as "the mother of the nation", that look set to help propel Park into the presidential Blue House.

Yuk is remembered for acts of charity that included a famous visit to a leper colony where she shook hands and embraced the sick. She remains South Korea's most popular first lady by far, polls show.

For many South Koreans, Park's frugal lifestyle as a single woman living in a modest home in the capital, Seoul, as well as her simple clothes and 1970s hairstyle bring her mother to mind.

"Park looks like her mother, when she greets people and smiles," one supporter, Lee Young-ho, told Reuters, sitting under the curved roof of Yuk's old home in the town of Okcheon, where she lived until she married Park.

Lee should know.

A former soldier and Vietnam War veteran, Lee was a member of the presidential guard in the 1970s and worked closely with the Park family.

The traditional Korean house where Lee spoke has become a shrine to Yuk with hundreds of people visiting every day.

Lee, now 68, is an official with Park's New Frontier Party and has also co-authored a biography of Yuk.

He is confident Park can maintain her double-digit lead in the polls and win an election victory which, he says, would somehow make up for the sacrifices.

"A kind of debt can be paid back when Park becomes president."

Yuk was 49 when, on August 15, 1974, she was killed by a stray bullet when a pro-North Korean assassin opened fire at her husband. Witnesses said the skies turning "reddish-purple" upon her death.

Memorial ceremonies are held on the anniversary at her grave in Seoul and in Okcheon, 170 km (100 miles) from the capital. Thousands of people attend.

"MOTHER'S DREAM"

Although her family's story is so well known, woven as it is into the fabric of the country's modern history, Park herself, who made two previous bids to win the conservative presidential nomination, is deeply private and cautious about her politics.

She disappeared from public life in 1979 after the assassination of her father by his intelligence chief, only to resurface in 1997 to "help save" the country from a devastating Asian financial crisis.

She has never clearly defined her policies on issues such as taxation, spending and welfare. She says she shares her mother's vision of a more equal society and has pledged to work for that.

"It was my mother's dream and her dream is now mine," she said at her mother's memorial service this month.

Critics say she has failed to apologize sufficiently for her father's rule.

She said her father's 1961 coup was "unavoidable and the best possible choice". At the same time, she says she has always been "apologetic" to victims of her father's brutal push for industrialization, when he crushed opposition in an effort to drive export-led growth.

Her likely opponent in the election, former human rights lawyer Moon Jae-in, who forged his political credentials in the pro-democracy movement, accuses Park of being a throwback to dictatorship.

"When I was living in poverty she was living the life of a princess in the Blue House," the left-of-center hopeful said when he declared his candidacy. "When I was fighting against dictatorship, she was at the heart of it."

Park, who shares her mother's hobby of embroidery and whose house is filled with pictures of her dead parents, according to a book about her, portrays her presidential bid as inspired by a sense of duty rather than privilege.

She once wrote that she might "choose death over a life like this again", referring to the killing of her parents.

It is mostly older people, those in their 50s and up, who attend the memorials for Yuk and form the bedrock of support for Park. Her weakness looks to be among younger, urban voters.

"I think Park has seen and learned a lot, going through the difficulties," said Lee Ae-joo, a nurse who was in the operating theatre when Yuk was brought in.

She recalls supplying blood to the wounded woman only to see it hemorrhage away. She also remembers Yuk's distraught husband staring at her body.

"I think Park has seen and learned a lot, going through the difficulties," said Lee, 66. "Right now, no one can compete with Park in the situation that our country is facing."

Lee is still impressed with Yuk's modesty, noting her underskirt looked as if it had been sewn by hand.

"The fabric looked like the kind of local product that you would get from the Dongdaemoon market," she said, referring to run-of-the-mill stalls in a bustling Seoul market where people hunt for bargains.

At the July rally at which Park launched her presidential bid, some people in the crowd held mementoes of the Park family.

"Honestly, Park Geun-hye is popular because of her mother's halo," said supporter Park Hong-pyo clutching a poem he had written dedicated to Yuk and old photographs of the Park family.

New Apple iPhone patent changes settings based on location ...

For those of you who may be Apple iPhone users or thinking of becoming we have some information about a new iPhone patent that changes settings based on location. We?re not about to claim that this may feature in the iPhone 5, expected to be unveiled in another couple of weeks, but it?s certainly a possibility at some point down the line for iPhone users and it?s interesting stuff indeed.

Some readers may already know that iOS 6, Apple?s next mobile operating system, is due to be released to the public next month and the iPhone 5 should launch running it. Regular readers may remember an article we wrote back in June about our wish list for iOS 6 and one of the features mentioned was for location-based profiles and actions that would enable your phone to automatically customize settings for different locations. Further to this we reported on news that iOS 6 could feature location check-ins via Yelp in the new Apple Maps app. It seems that Apple is already looking further ahead than this though and the patent could signal the future inclusion of dynamic location-aware settings.

A report on Apple Insider tells of a plethora of new Apple patents and one of these patents relates to wireless device ?enforcement policies.? This U.S. patent, number 8,254,902, listed as ?Apparatus and methods for enforcement of policies upon a wireless device,? would theoretically mean that customized alert profiles for various circumstances could be set up, to avoid for example those sticky situations where your iPhone rings in the funeral home or at the theater. Methods to enable these settings would be through WLAN communication, GPS and cellular triangulation to create a phone that would be ?situationally aware.?

The policy enforcement capability is described in the patent as, ?useful for a variety of reasons, including for example to disable noise and/or light emanating from wireless devices (such as at a movie theater), for preventing wireless devices from communicating with other wireless devices (such as in academic settings), and for forcing certain electronic devices to enter ?sleep mode? when entering a sensitive area.?

Various technical methods that would enable this are described at the Apple Insider link above and we think that the idea of having different profiles for different areas would no doubt appeal to many. In iOS 5 a limited version of this capability exists so that a reminder alert can be set for when you get to the office for example. However so far this is a limited function as well as being a battery-drainer. This new patent though goes several steps further and as well as being able to control different tones could also control brightness, radio communications and other features.

It?s certainly innovative stuff and we?d like to hear your thoughts on this new Apple patent and the possibility of location-based settings so send us your comments to let us know.

CAMBRIDGE, Mass. (August 28, 2012) The adult human circulatory system contains between 20 and 30 trillion red blood cells (RBCs), the precise size and number of which can vary from person to person. Some people may have fewer, but larger RBCs, while others may have a larger number of smaller RBCs. Although these differences in size and number may seem inconsequential, they raise an important question: Just what controls these characteristics of RBCs?

This question is particularly relevant for the roughly one-quarter of the population that suffers from anemia, which is often caused by flawed RBC production. A better understanding of how RBC production is controlled may offer greater insight into the development and potential treatment of anemia.

By analyzing the results of genome-wide association studies (GWAS) in conjunction with experiments on mouse and human red blood cells, researchers in the lab of Whitehead Institute Founding Member Harvey Lodish have identified the protein cyclin D3 as regulating the number of cell divisions RBC progenitors undergo, which ultimately affects the resulting size and quantity of RBCs. Their findings are reported in the September 14 issue of Genes and Development.

"This is one of the rare cases where we can explain a normal human-to-human variation," says Lodish, who is also a professor of biology and bioengineering at MIT. "In a sense, it's a window on human evolution. Why this should have happened, we have no idea, but it does."

Lodish likens cyclin D3's role in RBCs to that of a clock. In some people, the clock triggers RBC progenitors to mature after four rounds of cell division, resulting in fewer but larger RBCs. In others it goes off after five cell division cycles, which leads to production of a greater number of smaller RBCs. In both cases, the blood usually has the same ability to carry oxygen to distant tissues.

The initial hint of cyclin D3's importance came from GWAS, genetic surveys of large numbers of people with or without a particular trait. Researchers compare the groups in an attempt to identify genetic variations.

"The problem with most GWAS is that you get a bunch of potentially interesting genes, but that doesn't tell you anything about the functional biology, so you really have to figure it out," says Leif Ludwig, a Lodish graduate student and co-author of the Genes and Development paper. "You only know something has a role, but you don't know how it can cause variation. This work on cyclin D3 is a really nice example of how functional follow-up on a GWAS association can really teach us something about underlying biology."

In the case of RBC size and number, a mutation affecting cyclin D3 production bubbled to the surface from the GWAS's murky genetic data. Ludwig and co-author Vijay Sankaran then confirmed that reduced or inhibited cyclin D3 expression in mice and in human RBC progenitors caused those cells to halt cell division and mature earlier, producing larger and fewer red blood cells than mice and cells with uninhibited cyclin D3 production.

As one of only a handful of studies that have successfully used GWAS to produce definitive biological results, Sankaran is excited that this work confirms the value of such genetic studies.

"Can genetics teach us about biology?" asks Sankaran, also a postdoctoral researcher in the Lodish lab. "Yes! This work tells us that as genetic studies identify new genes, there will probably have been a lot of things biologists may have ignored. Genetics allows you to shine a spotlight on something interesting and then home in on it see what can be learned."

Wall Street ends flat, volume among lightest of year

NEW YORK (Reuters) - Stocks ended little changed in another day of scarce activity on Tuesday after mixed economic data gave investors little reason to shift their focus from Federal Reserve Chairman Ben Bernanke's speech on Friday.

The Fed chief is scheduled to address a conference of central bankers in Jackson Hole, Wyoming, and could announce new measures to boost growth. While Bernanke is expected to keep alive expectations for a third round of major bond buying by the Fed, or quantitative easing, he could keep markets guessing about the actual timing.

Volume was among the lightest of the year after Monday's lightest trading in 2012. August is a slow season, and investors mostly stayed on the sidelines, anticipating Bernanke's speech.

"I don't think you can read a lot from what's going on in the market right now," said John Fox, co-manager of the FAM Value Fund, in Cobleskill, New York. "Because there's nothing going on, everyone is waiting for the speech on Friday."

The latest sign of slowing in the global economy added to expectations for more central bank stimulus. Japan cut its assessment of economic growth, citing a deceleration in U.S. and Chinese demand for Japanese exports.

U.S. consumer confidence unexpectedly weakened in August to its lowest in nine months as Americans turned more pessimistic about the short-term outlook, according to the Conference Board.

But in another report, the S&P/Case Shiller composite index of 20 metropolitan areas showed U.S. home prices rose for a fifth consecutive month, a sign of slow improvement in the housing sector.

Among the day's biggest gainers, Lexmark International Inc jumped 13.7 percent to $21.62 after it said it would stop making inkjet printers, cut about 1,700 jobs, and focus on its more profitable imaging and software businesses.

The NYSEArca computer hardware index <.hwi> rose 1.2 percent. Shares of Hewlett Packard , which has a substantial portion of inkjet sales, were down 1.8 percent at $16.90.

The Dow Jones industrial average <.dji> was down 21.68 points, or 0.17 percent, at 13,102.99. The Standard & Poor's 500 Index <.spx> slipped 1.14 points, or 0.08 percent, at 1,409.30. The Nasdaq Composite Index <.ixic> edged down 3.95 points, or 0.13 percent, at 3,077.14.

Volume was 4.60 billion shares traded on the New York Stock Exchange, the Nasdaq and the NYSE MKT, compared with Monday's 2012 low 4.46 billion shares. The year-to-date average is about 6.6 billion.

The S&P 500 has been pinned in a fairly tight range over the last three weeks, finding a support level at 1,400 while also unable to convincingly pierce the April high of 1,422.38, which has acted as a resistance point. The index has been unable to muster a move of at least 1 percent in either direction since August 3.

Among other gainers, PVH Corp raised its full-year earnings outlook for the third time this year as it expects its European business to grow and its flagship Tommy Hilfiger brand to remain popular with shoppers. Shares climbed 4.8 percent to $92.77.

Movado Group Inc shares were up 17.4 percent at $35.36 after hitting an all-time high of $36.11. The watchmaker reported a higher second-quarter profit and raised its full-year earnings forecast for the second time.

Advancers outpaced decliners on the NYSE by about 4 to 3 and on the Nasdaq by about 14 to 9.

On Republican rival's big night, Santorum looks back

TAMPA, Florida (Reuters) - Looking back on the lessons of his own presidential campaign, when he was a nettlesome challenge to Mitt Romney, Rick Santorum returned to familiar themes of protecting traditional values in his address to the Republican National Convention on Tuesday.

Lingering tension between Santorum and Romney, who outlasted the former Pennsylvania senator in the Republican presidential primary, was clear in Santorum's sparing references to Romney. Santorum mentioned his former rival three times during a 14-minute address.

Santorum, a social conservative and vocal opponent of abortion rights, chose to focus on family, beginning with the inspiration of his coal miner grandfather and ending with that of his daughter Bella, who suffers from a severe genetic disorder.

Santorum recalled the many people who greeted him on the campaign trail.

"I grasped dirty hands with scars that come from years of labor in the oil and gas fields, mines and mills. Hands that power and build America and are stewards of the abundant resources that God has given us," Santorum said.

Santorum's repeated references to "hands" led David Axelrod, senior adviser to Democratic President Barack Obama to take to Twitter in reaction to Santorum's address: "All hands, no Mitt!"

The look back on his failed presidential campaign did not fit perfectly with the evening's forward-looking direction to a possible Romney presidency.

Santorum did echo a central theme of the Romney campaign, which has accused Obama of weakening work requirements in welfare.

The Obama administration's new rules give individual states more flexibility but do not remove the work requirement. Even so, Republicans have cast Obama's move as an assault on longstanding welfare-to-work programs.

As the Republican Party tried to focus the evening on the country's economic condition, Santorum closed with an appreciation of God and the Republican Party's dedication to pro-life causes.

Santorum's presence on the convention stage offered a counterpoint to Romney and a reminder of another possible course for the party.

2016?

An outspoken social conservative, Santorum proved a surprising obstacle to Romney's candidacy, bettering the former Massachusetts governor in 11 state primary contests.

That strong performance already has stirred questions of Santorum's intentions for the future and whether he would make a much-improved candidate in 2016 should Romney fall to President Barack Obama in the November 6 general election.

When politicians speak during conventions is a symbol of where they stand with the national party and, this year, with the Romney campaign.

The party appeared to pay deference to Santorum by placing him in the late evening hours. His slot was even moved back two hours later than scheduled.

Still, Santorum was kept out of the evening's brightest spotlight, which was reserved for Ann Romney and New Jersey Governor Chris Christie, another Republican frequently tipped as a possible 2016 candidate. Santorum took the podium before the three major television networks went live to the convention in prime time.

Romney's team allowed Santorum to speak at the convention after the former Pennsylvania senator allowed the Romney campaign to vet the speech.

Another former Romney rival, Texas Congressman Ron Paul, said he had declined to speak because he had refused to allow Romney's campaign to go over his remarks. But it's unclear whether Paul would have been given a slot anyway, because he has not endorsed Romney.

John Brabender, a senior strategist for Santorum, said Romney's campaign advisers read Santorum's speech "ahead of time, but they put no restrictions on us whatsoever, and they really had no changes whatsoever."

WARMING TO ROMNEY?

Since exiting the race on April 11, Santorum has kept a low-profile. He endorsed Romney in an email sent to supporters late at night in May ? a gesture seen as not entirely wholehearted.

The tentative embrace came after a contentious primary campaign where Santorum tagged Romney as the last person the party should nominate because of Romney's health care initiative in Massachusetts, a forerunner to the Obama plan.

This summer, Santorum has made rare appearances on Romney's behalf, but recently he has joined the campaign in challenging Obama's record on welfare.

Toward the end of his speech, Santorum praised Romney for his support of families and for backing the local control of schools, as well as lowering taxes.

On Wednesday, Santorum will hold a rally in support of Romney, gathering together conservative leaders like Tony Perkins of the Family Research Council, evangelical leader James Dobson, and former presidential candidate Gary Bauer.

Man apparently attempting Bigfoot hoax killed on Montana highway

MISSOULA, Montana (Reuters) - A man wearing a bushy, military-style camouflage suit in an apparent attempt to impersonate the mythical creature Bigfoot has died after being struck on a Montana highway by two cars, police said on Tuesday.

Randy Lee Tenley, 44, was dressed in a "ghillie" -- an outfit favored by military snipers and game hunters -- and standing in the middle of southbound lanes on U.S. Highway 93 near Kalispell on Sunday when he was struck twice in quick succession, Montana Highway Patrol spokesman Sergeant Steve Lavin said.

"From what I understand, at least one of his friends said that he was trying to induce a sasquatch sighting by using the suit along the highway," Lavin said. "This is a first for me after 20 years on the highway patrol. It's strange."

Tenley was first hit by a car driven by a 15-year-old girl (15-year-olds are allowed to drive in Montana with a learner's permit). A second car driven by a 17-year-old girl struck him moments later when he was already down on the roadway, Lavin said. Police said they do not know which impact killed him.

Friends of Tenley told Montana Highway Patrol trooper Jim Schneider that the man had attempted a similar hoax before, but never along the highway, police said.

Bigfoot, or sasquatch, is the name given to an ape-like creature whose existence has never been proven. Still, some people believe the creature lives in forest areas in North America, particularly the Pacific Northwest.

The Kalispell Army-Navy supply store sells the same kind of ghillie suit that police say Tenley was wearing.

Manager Dennis Petersen, who did not recall selling one to Tenley, said the shaggy $100 suits are often bought by boys who use them in paintball games. "I don't usually sell them to well-grown adults," he said.

It?s not uncommon when you browse the news sites to see yet another man suing a pharmaceutical manufacturer because the drug has caused erectile dysfunction. The most recent to hit the headlines has been a young man suffering from male pattern baldness who has discovered one of the problems in using the standard treatment. Given the warning given in the patient information leaflet sent out with the drug, the case looks unlikely to succeed, but this type of case is becoming more common. This is not to say there are a large number of drugs that do cause erectile dysfunction, but that those that may cause it are in quite common use. The best known has a complicated cause and effect. Many antidepressants affect libido but, equally, men who are depressed are less interested in sex. Nevertheless, it?s clear the erectile dysfunction will not be solved until the depression itself is treated. The erectile dysfunction drugs do not produce hard erections when men are seriously depressed. Men have to treat the depression before they can recover interest in sex.

However, there?s new research suggesting another possible long-term problem. A large number of men take painkillers over long periods of time. The classic example is using aspirin to keep a heart condition under control. But many take a painkiller every time they feel an ache. Not surprisingly, aches and pains become more common as we age so older men can find themselves taking one of the non-steroidal anti-inflammatory drugs (NSAIDs) five or six times a week. A new research paper published in the Journal of Urology shows a link between regular use of the routine painkillers and erectile dysfunction. Unfortunately, there are problems with the design of the study because there are no controls to filter out the contributions that might be made by lifestyle factors like eating a high-fat diet, drinking too much alcohol or refusing to quit smoking. It?s also well-established erectile affects men as they age with an increasing percentage of men over the age of 60 reporting the problem.

Nevertheless, the findings do challenge the orthodox assumption NSAIDs are not a threat, even over the long term. It will be interesting to see whether there?s further research to clarify the issue. A cynical view would be this research will be ignored. There?s billions of dollars earned through the sale of over-the-counter painkillers. If men got the idea it might be increasing the risk of erectile dysfunction as they aged, it could significantly affect sales. So we will probably not see much of a reference to this in our regular news media. When the pharmaceutical industry is such a big advertiser, it does not do to publish scare stories.

This leaves us with the problem of how to react to this research report. We could just say it?s a lone voice and until further research confirms it, we can safely continue with our regular use of painkillers. If erectile dysfunction does become a problem, Cialis will quickly restore hard erections so there?s no need to worry. Or we could say we take too many painkillers as a nation and this is a good reason to take less. If this saves us having to spend money of Cialis, we are ahead of the game.
